Chromatography separates and identifies components by distributing them between a stationary phase, which is usually packed in or attached to a supporting material, and a mobile phase that moves along or through the stationary phase.
When more than one component is present, bands are produced, which can be quantified in a chromatogram using a detector.
The detector response to parameters like solute concentration plotted against the elution time or volume produces a series of peaks called a chromatogram.
The time elapsed from the injection of the mixture to the detector&#39;s peak response to an analyte is the analyte&#39;s retention time, and the volume of the mobile phase required to elute the analyte is its retention volume.
The width at the base of a chromatographic peak, or bandwidth, is measured at the baseline between the tangents to the inflection points on the sides of the peak.
Another important concept is the peak resolution, measuring the separation of two adjacent peaks with respect to their peak widths at the base.
Additionally, the retention factor, or capacity factor, measures the analyte distribution between the stationary and mobile phases.&#160;

The liquid that flows out of the chromatography column goes past a detector, which creates a signal when it detects the solute being separated. This signal is then plotted against the elution time or the volume of solution eluted, resulting in a series of peaks known as a chromatogram.
In chromatography, several key terms help describe the process and results. The retention volume refers to the mobile phase required to carry an analyte from the injection point to the peak maximum in the chromatogram. Closely related is the retention time, which is the duration of time it takes for an analyte to travel through the system. This is calculated by dividing the retention volume by the flow rate of the mobile phase. Another important concept is the retention factor (often called the capacity factor), which measures the ratio of the time the analyte spends in the stationary phase to the time it spends in the mobile phase.&#160;
Additionally, void time indicates the time taken for unretained solutes to elute from the system. The corresponding void volume is the amount of mobile phase needed to elute these unretained solutes. When discussing the peaks in a chromatogram, the peak width, also known as the baseline width, is defined as the width of the peak at its baseline, measured where the peak starts to rise and where it returns to the baseline. Finally, resolution measures the degree of separation between two adjacent peaks, reflecting how well the components of a mixture are distinguished from one another.

Chromatography is an analytical technique used to separate the parts of a mixture and identify substances in it. It is used in chemistry, biology, environmental science, and pharmaceuticals. The method depends on two phases: the stationary phase, which stays fixed, and the mobile phase, which moves across it and carries the solutes.

As the mobile phase travels, each mixture component interacts with the phases in a different way. That difference causes the components to separate. Chromatography is grouped into several types based on the mobile and stationary phases. Common examples include gas chromatography (GC), liquid chromatography (LC), and thin-layer chromatography (TLC). Each type fits different materials and separation needs.

A detector sits after the chromatography column and measures the solute as it leaves the system. The detector makes a signal, and that signal is plotted against elution time or the volume of solution eluted. The result is a chromatogram, which shows a series of peaks. These peaks help show when different components pass through the system.
