Affinity chromatography is a separation technique to purify biomolecules from complex mixtures.
The separation is driven by a biospecific binding interaction between an analyte and its affinity ligand, such as antigens and antibodies, enzymes and substrates, or receptors and hormones.
The affinity ligand is in the stationary phase, being immobilized on a support such as agarose or porous glass beads, which are packed into a column.
The mixture containing the analyte is in the mobile phase, which must provide the right conditions for the binding interaction.
As the mixture passes through the column, the analyte selectively binds to the affinity ligand while the other molecules pass through the column.
Afterwards, the bound analyte is eluted by changing the mobile phase conditions, such as ionic strength or pH, to weaken the binding with the ligand and therefore obtaining the purified analyte.

The mobile phase serves a dual purpose &#8211; supporting strong binding between the analyte and ligand and subsequently weakening the interaction to elute the analyte. Only the analyte binds when the mixture containing the desired analyte is passed through the column, while other substances are washed through. The bound analyte can then be eluted by altering conditions like pH or ionic strength, which weakens its binding.
This method is particularly valuable in biochemistry for its specificity and application in isolating proteins, enzymes, substrates, antibodies, antigens, receptors, and hormones. Affinity chromatography can also be employed to isolate individual optical isomers of drugs, which may exhibit different therapeutic effects. This process involves using monoclonal antibodies specific to each stereoisomer.

Affinity chromatography separates and purifies specific biomolecules from a complex mixture. It depends on a highly selective match between an analyte and a binding partner, such as an antibody and an antigen. The binding partner is fixed on the stationary phase, which forms the affinity column.

The stationary phase is usually a solid support such as agarose or porous glass beads. The mobile phase helps the analyte bind to the ligand and later helps release it. When a sample passes through the column, only the target analyte binds. Other substances wash through without staying on the column.

The bound analyte is then eluted, or removed, by changing conditions such as pH or ionic strength. These changes weaken the interaction between the analyte and the ligand. This method is useful in biochemistry because of its high specificity.

Affinity chromatography is used to isolate proteins, enzymes, substrates, antibodies, antigens, receptors, and hormones. It can also separate individual optical isomers of drugs, which may have different therapeutic effects. In that case, monoclonal antibodies specific to each stereoisomer are used.
