Supercritical fluid chromatography, or SFC, is a hybrid technique with behavior between liquid and gas chromatography that uses a supercritical fluid&#8212;often carbon dioxide&#8212;as the mobile phase.
Supercritical fluids are formed by heating and pressurizing a substance to above its critical temperature and pressure.
The viscosity of a supercritical fluid is almost as low as that of its gas, while the density is close to that of its liquid. Its diffusion coefficient is between those of the gas and liquid.
So, SFC has faster and more efficient separation than LC does, but it&#39;s less affected by longitudinal diffusion than GC is. Importantly, it shares LC&#39;s compatibility with analytes that are difficult to volatilize.
SFC instruments are similar to standard HPLC instruments, except for a GC-like thermostatted column oven and a special pressure restrictor that maintain the mobile phase conditions.
SFC can use both open tubular and packed columns, which lets it employ detectors used with GC and HPLC, respectively. It is also easily coupled with MS and FTIR.

Supercritical fluid chromatography (SFC) provides a beneficial substitute for gas chromatography (GC) and liquid chromatography (LC) for certain samples because it merges the top attributes of both techniques. SFC allows the separation and analysis of compounds that GC or LC does not easily manage. These compounds are traditionally nonvolatile or thermally unstable, making GC unsuitable and lacking functional groups required for HPLC analysis.
SFC utilizes a supercritical fluid mobile phase, often CO2, which has properties between those of a gas and a liquid. The supercritical fluid&#39;s viscosity is the same as that of gases, while the density is closer to a liquid, and the diffusion coefficient is intermediate between gas and liquid. The low critical temperature and critical pressure of CO2 can be easily achieved and maintained.
SFC provides better analysis time and resolution than conventional HPLC. It is suitable for analyzing nonpolar organics and can also analyze polar solutes, including an organic modifier like methanol.
SFC shares similar instrumentation with GC and HPLC, with the addition of a pressure restrictor that maintains the critical pressure. Both packed and open tubular columns can be used as stationary phases in SFC. In SFC, packed columns offer a greater number of theoretical plates and can handle bigger sample volumes compared to open tubular columns. The low viscosity of supercritical media makes these columns longer than those found in LC. Open tubular columns resemble fused-silica wall-coated (FSWC) columns, while packed columns are commonly made of stainless steel. Initially, SFC utilized only polar stationary phases, but the growing use of modifiers has expanded the range of applicable phases. This makes it possible to modify retention mechanisms by adjusting the stationary and mobile phase compositions, integrating numerous HPLC stationary phases into SFC. SFC uses various detectors and allows for separating large molecules at relatively lower temperatures, resulting in increased efficiency and simplified coupling with MS or FT-IR.
Applications of SFC include the analysis of polymers, fossil fuels, waxes, drugs, and food products.

Supercritical fluid chromatography (SFC) is a separation method used for compounds that can be hard to analyze with gas chromatography (GC) or liquid chromatography (LC). It is useful for nonvolatile or thermally unstable samples that do not work well in GC and for compounds that do not have the right functional groups for HPLC analysis. SFC combines useful features of both GC and LC while keeping the analysis efficient.

SFC uses a supercritical fluid as the mobile phase, most often carbon dioxide (CO2). A supercritical fluid behaves like a gas in viscosity, like a liquid in density, and sits between the two in diffusion coefficient. The low critical temperature and critical pressure of CO2 make it practical to reach and hold these conditions during analysis. SFC can give better resolution and faster analysis times than conventional HPLC.

This method works well for nonpolar organic compounds and can also separate polar solutes when an organic modifier such as methanol is added. SFC uses instrumentation similar to GC and HPLC, but it includes a pressure restrictor to keep the critical pressure stable. Both packed columns and open tubular columns can serve as stationary phases. Packed columns provide more theoretical plates and can hold larger sample volumes, while the low viscosity of the supercritical mobile phase allows these columns to be longer than those used in LC.
