Flag Tag Immobilization

Flag Tag Immobilization is a biochemical technique that captures proteins engineered with a short FLAG epitope, enabling their selective attachment to a solid support for purification or analysis. The tagged protein binds an immobilized anti-FLAG antibody, typically on an affinity resin, while untagged molecules are removed during washing; the target can then be released using FLAG peptide or appropriate elution conditions. This method supports protein purification, pull-down assays, interaction studies, and biochemical characterization under controlled conditions. Because the tag is small and the antibody interaction is selective, Flag Tag Immobilization can isolate recombinant proteins while preserving access to their structure and activity.

Synthesis of custom plasmids is labor and time consuming. This protocol describes the use of Gibson assembly cloning to reduce the work and duration of custom DNA cloning procedure. The protocol described also produces reliable tagged protein constructs for mammalian expression at similar cost to the traditional cut-and-paste DNA cloning.
