Gpcr Beta-arrestin Interactions

GPCR beta-arrestin interactions are protein-protein associations that regulate how G protein-coupled receptors transmit and terminate signals, making them important in biochemistry and pharmacology. After ligand activation, GPCR kinases phosphorylate receptor sites, enabling beta-arrestin binding; this blocks further G protein coupling and can promote receptor internalization through clathrin-coated pits. Bound beta-arrestin can also serve as a scaffold for signaling pathways, including kinase cascades, so these interactions influence both signal duration and cellular responses. Studying them supports the design of biased ligands that selectively control G protein or beta-arrestin pathways, with potential to improve therapeutic efficacy and reduce adverse effects.
