Macroporous Hydrogels

Macroporous hydrogels are water-rich, three-dimensional polymer networks containing interconnected pores large enough to support fluid transport and, in some systems, cell or biomolecule movement. Their structure forms when polymer chains cross-link while a porogen, phase-separation process, or gas-forming reaction creates and preserves macropores within the swollen matrix. In biochemistry, these hydrogels provide accessible environments for enzyme immobilization, controlled molecular diffusion, and separation or purification processes. Their high water content, tunable pore architecture, and chemical functionality make them useful for studying biochemical interactions and developing platforms for tissue engineering, biosensing, and drug delivery.

Different methods to manipulate three-dimensional architecture in protein-based hydrogels are evaluated here with respect to material properties. The macroporous networks are functionalized with a cell-adhesive peptide, and their feasibility in cell culture is evaluated using two different model cell lines.
