Neonatal Cardiac Macrophages

Neonatal cardiac macrophages are tissue-resident and recruited immune cells in the newborn heart, where they connect innate immunity with cardiac development, repair, and homeostasis. They respond to local damage and biochemical signals, then use phagocytosis and efferocytosis to remove pathogens, debris, and dying cells while releasing cytokines and growth-associated factors that influence neighboring cardiomyocytes, fibroblasts, and vascular cells. Studying these macrophages helps explain why the neonatal heart can regenerate more effectively than the adult heart and may identify molecular pathways for reducing inflammation, supporting myocardial repair, and designing regenerative therapies for cardiac injury.

Primary mouse cardiomyocyte cultures are one of the pivotal tools for the investigation of myofibrillar organization and function. The following protocol describes the isolation and culture of primary cardiomyocytes from neonatal mouse hearts. The resulting cardiomyocyte cultures may be subsequently used for a variety of biomechanical, biochemical and cell-biological assays.
