Particle Trajectory Linking

Particle trajectory linking is a computational method that reconstructs the movement of individual particles by connecting their detected positions across successive image frames. It compares the locations of detections between frames and assigns them to the most plausible trajectories using criteria such as spatial proximity, expected motion, and continuity, while accounting for missed or newly appearing particles. In biochemistry, this approach supports analysis of single-molecule behavior, molecular transport, binding interactions, and diffusion in living systems or reconstituted assays. The resulting trajectories can reveal mobility states, reaction dynamics, and heterogeneity that ensemble measurements may obscure.

Particle-tracking microrheology investigates the viscoelasticity of materials. Here, the technique is used to determine the viscoelasticity, creep compliance and effective crosslinking roles of different matrix components of a bacterial biofilm. The matrix consists of polymeric substances secreted by the bacteria and its components determine biofilm structure and mechanical properties.

A method to analyze the solvent accessibility of the thiol group of cysteine residues of Maize rayado fino virus (MRFV)-virus-like particles (VLPs) followed by a peptide cross-linking reaction is described. The method takes advantage of the availability of several chemical groups on the surface of the VLPs that can be targets for specific reactions.
