Rab10 Thr73

Rab10 Thr73 is a specific threonine residue in the small GTPase Rab10 that serves as a key site of regulatory phosphorylation in cell signaling. The kinase LRRK2 phosphorylates Rab10 at Thr73, altering Rab10’s interactions with downstream effector proteins and influencing membrane trafficking processes controlled by the Rab GTPase cycle. Biochemical assays that measure Thr73 phosphorylation help characterize LRRK2 activity, substrate recognition, and pathway regulation. Because abnormal LRRK2 signaling is associated with Parkinson’s disease, Rab10 Thr73 is also an important molecular marker for studying disease mechanisms and evaluating kinase-targeted inhibitors.

Mutations in the leucine rich repeat kinase 2 gene (LRRK2) cause hereditary Parkinson’s disease. We have developed an easy and robust method for assessing LRRK2-controlled phosphorylation of Rab10 in human peripheral blood neutrophils. This may help identify individuals with increased LRRK2 kinase pathway activity.
