Synchrotron Instrument

A synchrotron instrument is a beamline-based system that uses radiation from a particle accelerator to probe matter with unusually high intensity, tunability, and spatial or temporal resolution. In a storage ring, relativistic electrons are guided by magnets, emitting synchrotron light that optics and monochromators select into an energy-defined beam before it reaches a sample and detector. In biochemistry, these instruments support macromolecular crystallography, small-angle X-ray scattering, and spectroscopy to reveal protein structures, conformational changes, and metal-site chemistry. The resulting measurements help explain molecular function, guide ligand studies, and evaluate structural changes under controlled experimental conditions.

Non-invasive 3D-Visualization with Sub-micron Resolution Using Synchrotron-X-ray-tomography

We used synchrotron X-ray tomography at the European Synchrotron Radiation Facility (ESRF) to non-invasively produce 3D tomographic datasets with a pixel-resolution of 0.7µm. Using volume rendering software, this allows the reconstruction of internal structures in their natural state without the artefacts produced by histological sectioning.

A molecular beam coupled to tunable vacuum ultraviolet photoionization mass spectrometer at a synchrotron provides a convenient tool to explore the electronic structure of isolated gas phase molecules and clusters. Proton transfer mechanisms in DNA base dimers were elucidated with this technique.

Using Synchrotron Radiation Microtomography to Investigate Multi-scale Three-dimensional Microelectronic Packages

For this study synchrotron radiation micro-tomography, a non-destructive three-dimensional imaging technique, is employed to investigate an entire microelectronic package with a cross-sectional area of 16 x 16 mm. Due to the synchrotron's high flux and brightness the sample was imaged in just 3 min with an 8.7 µm spatial resolution.
