Calcium Indicator Mice

Calcium indicator mice are genetically engineered animals that express fluorescent calcium sensors in selected cells, enabling researchers to monitor dynamic cellular activity in living tissues. When intracellular calcium binds to a genetically encoded indicator, the sensor changes fluorescence, allowing optical measurements of calcium signaling through microscopy or related imaging systems. In bioengineering, these models support studies of neuronal activity, muscle contraction, secretion, and other processes regulated by calcium. They also help evaluate engineered tissues, biomaterials, and therapeutic interventions by revealing how cells respond over time in physiologically relevant environments.

Loading Drosophila Nerve Terminals with Calcium Indicators

Calcium is a ubiquitous messenger in the nervous system, essential for triggering neurotransmitter release and changes in synaptic strength. Here we demonstrate a technique for loading Ca2+-indicators into Drosophila nerve terminals. We also demonstrate fabrication of the required apparatus and emphasize points critical for the technique's success.

This video analyzes T-cell receptor-induced calcium influx in T-cells. Anti-CD3 antibodies activate the T-cell signaling pathways, leading to an increase in intracellular calcium ions that bind with Indo-1 dye. In flow cytometry, a spectrum shift from the calcium-free state to the calcium-bound state correlates with T-cell receptor-induced calcium influx.

We describe methods to express genetically encoded calcium indicators in striatal astrocytes using adeno-associated viruses of serotype 2/5 (AAV2/5), as well as procedures to reliably image Ca2+ signals within striatal astrocytes in situ.
