Combinatorial Approach

A combinatorial approach is a strategy that systematically combines multiple components, variables, or design features to evaluate many possible solutions within a single investigation. In bioengineering, researchers create libraries of biomolecules, materials, genetic elements, or culture conditions and use parallel experimentation or high-throughput screening to identify combinations that produce a desired function. This method supports optimization of protein activity, biomaterial properties, metabolic pathways, and tissue-engineering environments while reducing reliance on trial-and-error testing. By linking controlled variation with quantitative analysis, combinatorial approaches reveal interactions among design factors and accelerate the development of improved biological systems and therapeutic technologies.

Bilayer Microfluidic Device for Combinatorial Plug Production

The fabrication of a polydimethylsiloxane (PDMS)-based bilayer device for the production of combinatorial libraries in water-in-oil emulsions (plugs) is presented here. The necessary hardware and software required to automate plug production are detailed in the protocol, and the production of a quantitative library of fluorescent plugs is also demonstrated.
