Cytotoxicity Reduction

Cytotoxicity reduction is the process of lowering the harmful effects that a material, compound, or engineered system can have on living cells, a key goal in bioengineering and biomedical design. It typically involves identifying toxic components, controlling exposure concentration and duration, and modifying material chemistry or surface properties to improve cellular compatibility while preserving function. Researchers assess outcomes through cell-viability, membrane-integrity, metabolic, or apoptosis assays, then use the results to refine formulations and device designs. These strategies support safer drug-delivery systems, biomaterials, implants, tissue-engineering scaffolds, and in vitro models, improving translational reliability and reducing adverse biological responses.

An Assay to Evaluate Immunodominance in Cytotoxic T Lymphocyte Responses

2025

The video showcases a flow cytometry-based assay for assaying immunodominance in cytotoxic T lymphocytes or CTL responses. Injected fluorescent target cells exhibit varying fluorescence corresponding to immunogenic epitopes. Post-spleen collection and sorting reveal a reduction in high-intensity fluorescent cells, confirming immunodominance in CTL responses.

Oxidation-Reduction Reactions

2020

Oxidation–Reduction Reactions Earth’s atmosphere contains about 20% molecular oxygen, O2, a chemically reactive gas that plays an essential role in the metabolism of aerobic organisms and in many environmental processes that shape the world. The term oxidation was originally used to describe chemical reactions involving O2, but its meaning has evolved to refer to a broad and important reaction class known as oxidation–reduction (redox) reactions. Some redox reactions involve the transfer of...

Evaluating Cytotoxicity and Membrane Integrity in Lung Slices via Lactate Dehydrogenase Assay

2025

This video demonstrates the lactate dehydrogenase, LDH assay to measure the cytotoxic effect of a test substance and its effect on the membrane integrity in human precision-cut lung slices. The assay provides a quantitative measure of cell death or damage in the sample.

An Immunological Technique to Monitor Adjuvant-Mediated Cytotoxic T Lymphocyte Generation

2025

The video demonstrates an immunological method to monitor in vivo adjuvant-induced cytotoxic T lymphocyte or CTL production. A mouse is pre-injected with genetically engineered, proliferative dye-stained donor CD8+ T lymphocytes. Subcutaneous injection of ovalbumin with adjuvants generates cytotoxic CD8+ T lymphocytes that are identified post-harvesting through immunostaining and flow cytometry.
