Dynamic Force Spectrum

Dynamic Force Spectrum is a bioengineering framework for characterizing how mechanical forces and force responses vary with time, loading rate, or frequency. In practice, a controlled time-varying load is applied to a biological material, cell, tissue, or molecular interaction, and the resulting force response is recorded to reveal features such as viscoelastic behavior, adhesion, deformation, or bond dissociation. Comparing responses across loading conditions produces a force spectrum that links mechanical measurements to underlying structure and kinetics. This approach supports the study of cell-material interactions, biomolecular binding, tissue mechanics, and the design and evaluation of engineered biomaterials and medical devices.

In this video, we demonstrate high-speed time-lapse atomic force microscopy, AFM, imaging techniques to study the dynamics of nucleosome complexes. As the AFM tip scans across the surface of the nucleosome, it detects alterations in the nucleosome height and records the DNA unwrapping over time.
