Membrane Fluidity Assay

A membrane fluidity assay is a method for measuring how freely lipids and membrane-associated molecules move within a biological or engineered membrane, a property that influences membrane stability, signaling, and transport. The assay commonly uses fluorescent probes to monitor changes in fluorescence anisotropy or recovery after photobleaching, revealing lateral diffusion under defined conditions such as altered temperature, lipid composition, or cholesterol content. In bioengineering, these measurements help characterize cell membranes, optimize liposomes and other biomimetic membranes, and evaluate materials for drug delivery and biosensors. Quantifying fluidity also supports the design of membrane-based systems with controlled permeability, mechanical behavior, and molecular organization.

An In Vitro Fluorescence-Based Assay to Measure Plasma Membrane Resealing Efficiency

This video demonstrates an assay to study the plasma membrane resealing efficiency in mammalian cells. The cells are exposed to a bacterial pore-forming toxin, which forms pores in the cell membrane. In the presence of extracellular calcium ions, the pore causes calcium influx, inducing membrane-resealing events. The resealing efficiency is assessed using DNA-binding propidium iodide dye, which only enters cells without a resealed membrane.

Testing protein-protein interaction is indispensable for dissection of protein functionality. Here, we introduce an in vitro protein-protein binding assay to probe a membrane-immobilized protein with a soluble protein. This assay provides a reliable method to test interaction between an insoluble protein and a protein in solution.
