Multiplexed Analyte Detection

Multiplexed analyte detection is the simultaneous identification and measurement of multiple chemical or biological targets in a single sample, increasing information while conserving sample volume and time. It works by pairing each analyte with a selective recognition element, such as an antibody, nucleic-acid probe, or engineered binding molecule, and assigning the interaction a distinguishable signal, location, or code; these signals are then resolved and quantified together. In bioengineering, multiplexed assays support biomarker profiling, pathogen characterization, cell analysis, and monitoring of complex biological systems. Advances in panel design, signal separation, and computational analysis continue to expand their value in diagnostics and research.

Multiplex fluorescence immunoassays are becoming a reference for serology testing. Their application, however, is limited by the complexity and time-consuming bead-analyte coupling processes. This report presents a semi-automated, reproducible multiplex microsphere bead coupling protocol that is comparable to the standard method while considerably reducing time to results.
