Refractive Index Matching Solution

A refractive index matching solution is a liquid medium formulated to approximate the refractive index of a biological sample and its surrounding environment, reducing optical distortion and light scattering. By minimizing differences in refractive index at tissue interfaces, the solution improves light transmission and enables clearer imaging through specimens, materials, or microfluidic systems. In bioengineering, these solutions support microscopy, tissue imaging, biomaterial characterization, and visualization of engineered constructs, helping researchers assess structure and function with greater accuracy. Their use can improve image quality without requiring extensive changes to optical hardware, making them valuable for quantitative analysis and the development of advanced diagnostic and tissue-engineering methods.

This protocol describes the fabrication of microfluidic devices from MY133-V2000 to eliminate artifacts that often arise in microchannels due to the mismatching refractive indices between microchannel structures and an aqueous solution. This protocol uses an acrylic holder to compress the encapsulated device, improving adhesion both chemically and mechanically.

Reflection and Refraction

Source: Derek Wilson, Asantha Cooray, PhD, Department of Physics & Astronomy, School of Physical Sciences, University of California, Irvine, CA Light travels at different speeds depending on the material through which it is propagating. When light travels from one material to another, it will either slow down or speed up. In order to conserve energy and momentum, the light must change the direction in which it propagates. This bending of light is known as refraction. Some fraction of the...

Sign Test for Matched Pairs

2025

The sign test for matched pairs offers a robust method for comparing two paired samples, often for the effects of an intervention in one of them. This method is very useful in situations where the underlying distribution of the data is unknown. The test compares two related samples—often pre- and post-treatment measurements on the same subjects—to determine if there are significant differences in their median values. To conduct the sign test, we first calculate the differences in value between...

Making Solutions in the Laboratory

2023

The ability to successfully make solutions is a basic laboratory skill performed in virtually all biological and chemical experiments. A solution is a homogenous mixture of solute dissolved in bulk liquid known as the solvent. Solutions can be described by their solute concentration, a measure of how much solute is present per unit of solution. In this video, a step-by-step procedure for how to make a water-based, or aqueous, solution for biological applications is presented. The video...
