Endothelial Cell Engineering

Endothelial cell engineering is the design, modification, and cultivation of endothelial cells to control how they form and function as the inner lining of blood vessels. In bioengineering, researchers combine cell culture with biochemical signals, biomaterials, and physical cues such as fluid shear stress to promote cell adhesion, alignment, barrier formation, and vascular network development. These approaches support the creation of engineered blood vessels and vascularized tissues, while providing models for studying angiogenesis, inflammation, and vascular disease. By improving control over vascular behavior, endothelial cell engineering contributes to tissue regeneration, drug testing, and the development of more physiologically relevant laboratory systems.

Isolation of Valvular Endothelial Cells

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.

Protein transduction enables the direct delivery of biologically active proteins into cells. In contrast to conventional methods such as DNA transfection or viral transduction this non-invasive paradigm allows highly efficient cellular manipulation in a titratable manner circumventing cellular toxicity and the risk of oncogenic transformation by permanent genetic modification.

Isolation of Endothelial Progenitor Cells from Human Umbilical Cord Blood

The goal of this protocol is to isolate endothelial progenitor cells from umbilical cord blood. Some of the applications include using these cells as a biomarker for identifying patients with cardiovascular risk, treating ischemic diseases, and creating tissue-engineered vascular and heart valve constructs.

Isolation of Murine Embryonic Hemogenic Endothelial Cells

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.
