Antibody Subtype Detection

Antibody subtype detection is the identification of an antibody’s class, subclass, or isotype, such as IgG, IgM, IgA, or IgE, and is important for characterizing immune responses and evaluating antibody-based assays. The method typically uses subtype-specific antibodies or binding reagents that recognize distinct constant-region sequences, producing a measurable signal through techniques such as enzyme-linked immunosorbent assays, immunoblotting, or flow cytometry. In biological research, subtype detection helps distinguish antigen-specific responses, assess antibody production, confirm reagent identity, and select appropriate antibodies for diagnostic, therapeutic, and experimental applications.

We describe a set of assays to analyze expression levels of H1 linker histones. mRNA of individual H1 genes are quantitatively measured by random primer based reverse transcription followed by real-time PCR, whereas protein quantification of H1 histones is achieved by HPLC analysis.

Use of an Influenza Antigen Microarray to Measure the Breadth of Serum Antibodies Across Virus Subtypes

We present a protocol for using a protein microarray constructed by printing influenza antigens onto nitrocellulose-coated slides to simultaneously probe serum for multiple antibody isotypes against over 250 antigens from different virus strains, thus allowing measurement of the breadth of serum antibodies across virus subtypes.
