6-bisphosphate

Fructose-1,6-bisphosphate is a six-carbon sugar phosphate and a central intermediate in glycolysis, the pathway that converts glucose into usable cellular energy. During glycolysis, phosphofructokinase-1 adds a second phosphate group to fructose-6-phosphate, producing fructose-1,6-bisphosphate and committing the molecule to metabolic breakdown. Aldolase then cleaves it into the three-carbon intermediates glyceraldehyde-3-phosphate and dihydroxyacetone phosphate, which continue through the pathway to generate ATP and NADH. Because its formation regulates glucose catabolism, fructose-1,6-bisphosphate helps coordinate energy production with cellular demands and provides a key framework for studying metabolic regulation in biology.

PI(4,5)P2 regulates various cellular functions, but its nanoscale organization in the cell plasma membrane is poorly understood. By labeling PI(4,5)P2 with a dual-color fluorescent probe fused to the Pleckstrin Homology domain, we describe a novel approach to study the PI(4,5)P2 spatial distribution in the plasma membrane at the nanometer scale.

Generating the 6-hydroxydopamine Rat Model of Parkinson's Disease

This video demonstrates a procedure to create a rat model of Parkinson's disease by injecting the neurotoxin 6-hydroxydopamine (6-OHDA) into the brain. The neurotoxin infusion into the medial forebrain bundle (MFB) specifically targets dopaminergic neurons responsible for motor control, leading to their destruction and replicating Parkinson's disease symptoms.

To model how inflammatory signals influence neurodevelopment in genetically susceptible organisms, we present a protocol for microinjecting IL-6 into the duct of Cuvier (common cardinal vein) of zebrafish larvae, enabling controlled induction of inflammation and assessment of its developmental impact in the context of tuberous sclerosis complex (TSC).

The 6-hydroxydopamine (6-OHDA) model has been used for decades to advance the understanding of Parkinson's Disease. In this protocol, we demonstrate how to perform unilateral nigrostriatal lesions in the rat by injecting 6-OHDA in the medial forebrain bundle, assess motor deficits, and predict lesions using the stepping test.
