Activator Proteins

Activator proteins are regulatory molecules that increase the expression of specific genes, making them essential for controlling cellular identity, development, and responses to environmental signals. Many activators function as transcription factors: they recognize regulatory DNA sequences such as enhancers or promoter regions and recruit coactivators, chromatin-remodeling complexes, or RNA polymerase to promote transcription. Their activity can be controlled by signaling pathways, protein modification, ligand binding, or interactions with other regulatory proteins. Studying activator proteins helps explain gene regulation in normal and diseased cells and supports applications in developmental biology, disease research, biotechnology, and the design of targeted genetic interventions.

Protein kinases are highly evolved signaling enzymes and scaffolds that are critical for inter- and intracellular signal transduction. We present a protocol for measuring kinase activity through the use of radiolabeled adenosine triphosphate ([γ-32P] ATP), a reliable method to aid in elucidation of cellular signaling regulation.

Quantifying Agonist Activity at G Protein-coupled Receptors

A method for estimating the affinity constant of an agonist for the active state (Kb) of a G protein-coupled receptor is described. The analysis provides absolute or relative measures of Kb depending on whether constitutive receptor activation is measurable. Our method applies to various responses downstream from receptor activation.
