Adult Topical Assay

An adult topical assay is a biological method for evaluating how a substance affects mature organisms after researchers apply a measured dose directly to the external surface. In insect studies, the compound penetrates the cuticle and enters the body, allowing investigators to relate exposure dose to outcomes such as mortality, immobilization, or other physiological responses under controlled conditions. This assay supports dose-response analysis, comparison of insecticidal activity, and screening of candidate compounds while reducing variation associated with oral or environmental exposure. Results can inform toxicology, pest-management research, and the development of more selective control strategies.

In this video, we demonstrate the topical testing assay to determine the toxicity of a potential insecticide on adult female mosquitoes. Using a dopamine receptor antagonist that causes lethal inhibition of neurotransmission in mosquitoes, this method describes a way to evaluate the toxic effects of insecticides upon topical application.

An Assay for Lateral Line Regeneration in Adult Zebrafish

Because many zebrafish models of neurological and non-neurological diseases are studied in the adult fish rather than the embryo/larvae, we developed a quantitative lateral line regenerative assay that can be applied to adult zebrafish disease models. The assay involved resolution at the 1) neuromast and 2) individual hair cell levels.

Generation of Topically Transgenic Rats by In utero Electroporation and In vivo Bioluminescence Screening

Genes can be manipulated during development of the cortex or the hippocampus of the rat via in utero electroporation (IUE) at E16, to enable fast and targeted modifications in neuronal connectivity for later studies of behavior or neuropathology in adult animals. Postnatal in vivo imaging for control of IUE success is performed by bioluminescence of activating co-transfected luciferase.
