Agrobacterium Mediated Transformation

Agrobacterium-mediated transformation is a method for introducing foreign DNA into plant cells using the natural genetic engineering ability of Agrobacterium tumefaciens. When the bacterium encounters wounded plant tissue, virulence genes activate the transfer of T-DNA from the Ti plasmid into the plant cell, where the DNA can enter the nucleus and integrate into the plant genome. Researchers use modified T-DNA vectors to introduce genes while removing disease-causing sequences, enabling stable production of transgenic plants. This technique supports functional genomics, crop improvement, disease resistance studies, and the development of plants with valuable agricultural or biotechnological traits.

Gene deletion mutants generated through homologous recombination are the gold standard for gene function studies. The OSCAR (One Step Construction of Agrobacterium-Recombination-ready-plasmids) method for rapid generation of deletion constructs is described. Agrobacterium mediated fungal transformation follows. Finally, a PCR based confirmation method of gene deletions in fungal transformants is presented.

We present an efficient, optimized protocol for Agrobacterium-mediated transformation of an asexual amphibious Brassicaceae species, Rorippa aquatica, without resource-intensive sterile tissue culture procedures. Utilizing a visual transformation marker and shoot regeneration pipeline successfully generated near clonal transformants, thereby advancing the future studies of plant's adaptation to the underwater environment.

Agrobacterium tumefaciens-Mediated Genetic Transformation of Narrowleaf Plantain

Because of its versatile application as a model species in various fields of study, there is a need for a genetic transformation toolkit in narrowleaf plantain (Plantago lanceolata). Here, using Agrobacterium tumefaciens-mediated transformation, a protocol is presented that results in stable transgenic lines with a transformation efficiency of 20%.
