Macrophage Endothelial Co-culture

Macrophage endothelial co-culture is an in vitro model that grows macrophages and endothelial cells together to study how immune and vascular cells influence one another. Through direct cell contact and soluble signals released into the shared culture environment, macrophages can alter endothelial activation, barrier behavior, and inflammatory responses, while endothelial cells regulate macrophage phenotype and function. Researchers use this system to model vascular inflammation, immune cell recruitment, angiogenesis, and tissue repair under controlled laboratory conditions. Compared with single-cell-type cultures, the model captures multicellular communication more realistically, supporting investigation of disease mechanisms and evaluation of therapeutic strategies targeting vascular or inflammatory pathways.

Assessment of Leishmania Virulence within Cultured Macrophages

2025

This video investigates Leishmania's virulence by treating macrophages with metacyclic promastigotes, resulting in the formation of intracellular amastigotes. The confirmation of Leishmania's virulence is achieved through nucleic acid fluorescent dye staining. This insight can contribute to the development of new therapies for leishmaniasis.

Culture of Macrophage Colony-stimulating Factor Differentiated Human Monocyte-derived Macrophages

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.

In Vitro Culture and Differentiation of Primary Monocytes into Macrophages

This video demonstrates the priming of primary monocytes into macrophages. Primary human monocytes can be isolated and differentiated to an M1-like pro-inflammatory phenotype using the granulocyte-macrophage colony-stimulating factor (GM-CSF) or to an M2-like anti-inflammatory phenotype using the macrophage colony-stimulating factor (M-CSF).

Establishing a Co-Culture of Interneurons with Periventricular Endothelial Cells

2025

This video demonstrates a technique for establishing a co-culture of interneurons with perivascular endothelial cells. The cells are added to a one-well insert placed inside a culture dish. After the cells adhere to the dish, the insert is removed, allowing the cells to proliferate and interact and form a co-culture.
