Alkaline Tide

Alkaline tide is the temporary increase in blood bicarbonate and systemic alkalinity that follows gastric acid secretion after a meal, making it an important example of coordinated acid–base physiology. In gastric parietal cells, carbonic anhydrase generates hydrogen ions and bicarbonate from carbon dioxide and water; hydrogen ions enter the stomach lumen to form hydrochloric acid, while bicarbonate moves into the bloodstream, and the kidneys subsequently excrete excess bicarbonate in more alkaline urine. Studying alkaline tide helps explain normal digestion, renal acid–base handling, and disturbances such as prolonged vomiting, while linking gastrointestinal activity with whole-body pH regulation.

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.
