Background Flattening Filter

A Background Flattening Filter is an image-processing tool that reduces gradual variations in brightness so biological features stand out more clearly against uneven backgrounds. It estimates the low-frequency background intensity, often by smoothing the image or applying a morphological operation, and then subtracts or normalizes that signal while preserving sharper structures such as cells, tissue boundaries, or fluorescent objects. In microscopy, this correction can improve contrast, segmentation, and quantitative measurements when illumination, staining, or optical conditions produce intensity gradients. Background flattening supports more reliable visualization and analysis of complex biological samples without altering the underlying specimen.

Source: Lauer, S. M., et al. Visualization of Cortical Modules in Flattened Mammalian Cortices. J. Vis. Exp. (2018) This video demonstrates a method to visualize the cortical modules in flattened brain sections using histochemical staining. Pre-fixed sections are treated with a straining reagent that highlights active neural clusters, forming dark, high-contrast regions. After mounting and dehydration, cortical modules appear under a bright-field microscope as dark, patchy patterns.

The law of mass action states that "the rate of a chemical reaction is directly proportional to the product of the molar concentrations of the reactants." It means that the more 'active mass' or 'concentration' of the reactants present, the faster the reaction will proceed. In a chemical reaction, there are forward and reverse reactions. The forward reaction is the process where the reactants combine to form products. The reverse reaction is the process where the products break down to form the...

Passive filters are utilized to shape the frequency spectrum of signals across a diverse array of applications. These filters, using only passive elements like resistors (R), inductors (L), and capacitors (C), are capable of selectively allowing or blocking certain frequency ranges without the need for external power sources. Low-Pass Filters Low-pass filters are designed to transmit signals with frequencies lower than the cutoff frequency, ωc, and attenuate those above it. The cutoff frequency...

We discuss the construction and operation of a complex nonlinear optical system that uses ultrafast all-optical switching to isolate Raman from fluorescence signals. Using this system we are able to successfully separate Raman and fluorescence signals utilizing pulse energies and average powers that remain biologically safe.
