Bacterial Colony Miniprep

Bacterial colony miniprep is a small-scale molecular biology method for isolating plasmid DNA from bacteria, making it possible to identify and analyze recombinant clones efficiently. The procedure typically resuspends a bacterial colony, disrupts cells with an alkaline detergent solution, and then neutralizes the lysate so genomic DNA, proteins, and cell debris can be separated from soluble plasmid DNA, often by centrifugation and silica-column purification. The recovered plasmids can be evaluated by restriction digestion, PCR, or sequencing and used for downstream cloning, expression studies, and confirmation of genetic constructs in biology research.
