BCECF is a fluorescent pH indicator used to measure intracellular acidity in living cells, making it valuable for studying ion regulation and cellular physiology. The membrane-permeable BCECF-AM form enters cells, where esterases remove the acetyl groups and retain the dye; protonation changes its fluorescence, allowing pH estimation through intensity or excitation-wavelength ratios. Researchers use BCECF to monitor cytoplasmic pH, characterize proton transport, and assess how cells respond to metabolic, environmental, or pharmacological challenges. These measurements help connect membrane activity and cellular stress with processes such as signaling, homeostasis, and disease-related dysfunction.

Visualization of Internalized Anaerobic Bacteria within Host Endothelial Cells

In this video, we demonstrate a procedure to visualize internalized Porphyromonas gingivalis, an anaerobic pathogenic bacterium, within human endothelial cells using confocal microscopy. The bacteria are stained with a fluorescent dye while the host endothelial cell actin cytoskeleton is labeled with a different fluorescent dye. The internalized Porphyromonas gingivalis inside endothelial cells are visualized using confocal microscopy.
