Biuret Test

The Biuret Test is a chemical assay used to detect proteins and estimate their concentration, making it a foundational method in biology and biochemistry. In an alkaline solution, copper(II) ions react with peptide bonds in proteins to form a violet-colored coordination complex; the color intensity generally increases with the amount of protein present and can be measured visually or with a spectrophotometer. Researchers and students use this test to identify protein-containing samples, compare relative protein levels, and monitor protein purification or experimental treatments. Because the reaction depends on peptide bonds, free amino acids typically produce little or no positive result.

This method aims at locating vertical subsurface defects. Here, we couple a laser with a spatial light modulator and trigger its video input to heat a sample surface deterministically with two anti-phased modulated lines while acquiring highly resolved thermal images. The defect position is retrieved from evaluating thermal wave interference minima.
