Calcium Handling Assessment

Calcium handling assessment is the evaluation of how cells regulate calcium ions, a process that supports signaling, contraction, secretion, metabolism, and other biological functions. It examines changes in intracellular calcium as ions enter through channels, are released from internal stores, bind cellular targets, and are removed or sequestered to restore baseline conditions. In biology, these assessments help characterize calcium dynamics in muscle, nerve, and other excitable or signaling cells, identify disruptions in cellular regulation, and compare responses to physiological or experimental stimuli. The resulting measurements provide insight into cell function and mechanisms underlying disease.

Anti-CD3 antibodies activate the T-cell signaling pathways, leading to an increase in intracellular calcium ions that bind with Indo-1 dye. In flow cytometry, a spectrum shift from the calcium-free state to the calcium-bound state correlates with T-cell receptor-induced calcium influx.

Intracellular calcium recycling plays a critical role in regulation of systolic and diastolic function in cardiomyocytes. Here, we describe a protocol to evaluate sarcoplasmic reticulum Ca2+ reserve and diastolic calcium removal function in cardiomyocytes by a calcium imaging system.
