Ccr7-negative T Cells

Ccr7-negative T cells are T lymphocytes that lack detectable expression of CCR7, a chemokine receptor that helps direct immune cells into lymphoid tissues. Because they do not respond efficiently to the CCR7 ligands CCL19 and CCL21, these cells show reduced lymph-node homing and are commonly associated with migration toward peripheral tissues and effector-memory immune responses. Identifying Ccr7-negative T cells by flow cytometry or other immunophenotyping methods helps researchers distinguish T-cell subsets, track activation and differentiation, and investigate immune responses in infection, cancer, autoimmunity, and tissue inflammation.

DNabT cells are rare among peripheral T cells; however, they are abundant in certain non-lymphoid tissues. Difficulty of isolating DN T cells from non-lymphoid tissue hinders their functional analysis despite increasing recognized pathophysiologic significance. We describe a novel protocol for isolation of highly purified DN T cells from murine kidney.

The exposed normal ocular surface consists of cornea and conjunctiva. Epithelial cells, goblet cells and immune cells are present in the conjunctiva. Here, a non-invasive, technique of impression cytology is described using an impression cytology device and flow cytometry to analyze immune cells in the conjunctiva.

Negative Immunomagnetic Selection: A Method to Purify B-cells from Peripheral Blood Mononuclear Cells

2023

The video explains a way to purify B-cells by immunomagnetic targeting and separation from peripheral blood mononuclear cells suspension. The method involve labeling unwanted cell types with specific antibodies or ligands (linked to magnetic particles) targeting specific cell surface proteins, to be depleted from the heterogenous cell suspension by magnetic separation.

Melanoma Stem Cell Isolation: A Method to Obtain CD133 Positive and Negative Melanoma Cells Using Magnetic-activated Cell Sorting

2023

This video describes the technique of isolating CD133 negative and positive melanoma stem cells from a primary melanoma cell line using magnetic-activated cell sorting. The CD133 positive melanoma CSCs are further studied to understand the mechanisms of melanoma tumorigenesis.
