Confocal Microscopy

Confocal microscopy is an optical imaging technique that produces sharp, high-resolution images by eliminating light from regions outside the focal plane, making it valuable for examining biological structures in three dimensions. A focused laser scans fluorescently labeled specimens point by point, while a detector positioned behind a pinhole blocks out-of-focus emission; collecting sequential optical sections through the sample enables reconstruction of a three-dimensional image. In biology, this approach reveals the spatial organization of cells, tissues, and subcellular components, supports analysis of protein localization and cell morphology, and allows researchers to study dynamic processes in living specimens when compatible fluorescent probes and imaging conditions are used.

Human sclera tissue is mainly collagen; therefore, it is not easily usable for immunohistochemistry. To achieve the goal of performing immunohistochemistry for confocal microscopy of scleral tissue, a laminating technique was used.
