Cross-beta Filament

Cross-beta filaments are elongated protein assemblies built from beta-sheet structures, a defining architecture of amyloid fibers and other ordered protein aggregates. In this arrangement, beta strands lie roughly perpendicular to the filament axis while hydrogen bonds stabilize sheets that extend along the axis, producing a strong, repetitive molecular scaffold. Cross-beta structure helps explain the persistence and mechanical stability of amyloid deposits associated with neurodegenerative and systemic diseases. Its identification through methods such as X-ray fiber diffraction, cryo-electron microscopy, and spectroscopy supports studies of protein misfolding, disease mechanisms, biomolecular assembly, and the design of functional protein-based materials.
