Death Receptors

Death receptors are cell-surface receptors that regulate programmed cell death and other responses to extracellular signals, making them important for tissue homeostasis and immune function. Most belong to the tumor necrosis factor receptor superfamily and contain cytoplasmic death domains; when ligands such as Fas ligand or tumor necrosis factor bind, the receptors recruit adaptor proteins that assemble signaling complexes and activate initiator caspases, particularly caspase-8, to trigger apoptosis. Death receptor pathways also interact with inflammatory signaling and survival mechanisms. Their study helps explain immune-cell regulation, developmental processes, autoimmune disease, and cancer, where altered receptor signaling can promote unwanted cell loss or resistance to therapy.

This video demonstrates an ex vivo technique to induce brain cell death mediated through cytotoxic CD8+ T cells. Upon obtaining brain slices, activated oligodendrocyte-directed CD8+ T cells are transferred onto the slices to target the myelin sheath and oligodendrocytes.
