Dextran Permeability Assay

The dextran permeability assay is a method for measuring how readily dextran molecules cross a biological barrier, providing an indicator of barrier integrity and paracellular transport. In a typical experiment, a fluorescently labeled dextran is added to one side of a cell layer or membrane, and its movement to the opposite side is quantified over time; larger dextrans generally cross less readily when tight junctions and cell-cell contacts remain intact. In biology, this assay is used to evaluate endothelial and epithelial permeability, monitor changes caused by inflammation or chemical treatment, and compare barrier function in disease models, tissue engineering, and drug research.

We describe a live whole animal quantitative measurement for permeability of the embryonic zebrafish brain. The technique analyzes the ability to retain cerebrospinal fluid and molecules of different molecular weights within the neural tube lumen and quantifies their movement out of the ventricles. This method is useful for determining differences in epithelial permeability and maturation during development and disease.
