Disease Mechanisms

Disease mechanisms are the biological processes through which genetic, environmental, or infectious factors disrupt normal structure and function, producing illness. They may involve altered gene expression, abnormal cell signaling, inflammation, immune responses, metabolic imbalance, or pathogen-driven damage that progressively changes tissues and organs. Studying these mechanisms connects molecular events to clinical symptoms and disease progression, helping researchers identify biomarkers, classify disease subtypes, and develop targeted treatments. In biology, mechanism-based analysis also supports experimental models, therapeutic testing, and prevention strategies by revealing how disturbances arise, interact, and potentially be reversed.

Lipoxygenase (LOX) isozymes can generate products that may increase or decrease neuroinflammation and neurodegeneration. A gene-environment interaction study could identify LOX isozyme-specific effects. Using the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) model of nigrostriatal damage in two LOX isozyme-deficient transgenic lines allows for comparison of the contribution of LOX isozymes on dopaminergic integrity and inflammation.

The biosynthesis of cartilaginous extracellular matrix by chondrocytes can be affected by application of mechanical stimuli. This method describes the technique of applying dynamic compressive strains to chondrocytes encapsulated in 3D constructs and the evaluation of induced changes in chondrocyte metabolism.

Chemical reactions often occur in a stepwise fashion, involving two or more distinct reactions taking place in a sequence. A balanced equation indicates the reacting species and the product species, but it reveals no details about how the reaction occurs at the molecular level. The present protocol describes the development of a Crohn's-like colitis model in rodents. Transmural inflammation leads to stenosis at the TNBS instillation site, and mechanical enlargement is observed in the segment proximal to the stenosis. These changes allow studying mechanical stress in colitis.
