Dna Binding Dye

A DNA-binding dye is a chemical probe that associates with DNA, allowing the molecule to be detected, visualized, or quantified in biological samples. These dyes typically bind by intercalating between base pairs or fitting within the DNA minor groove, which can alter their absorption or increase their fluorescence; the resulting signal depends on dye concentration and DNA binding. In biology, DNA-binding dyes support nucleic acid staining in gels, fluorescence microscopy, and DNA quantification assays. They are also used to monitor DNA amplification, assess sample quality, and analyze cellular or molecular processes involving changes in DNA content.

Here we present a simple and efficient method to isolate live meiotic and post-meiotic germ cells from adult mouse testes. Using a low-cytotoxicity, violet-excited DNA binding dye and fluorescence-activated cell sorting, one can isolate highly enriched spermatogenic cell populations for many downstream applications.
