Drosophila Screening

Drosophila screening is a genetic and phenotypic approach that uses fruit flies to identify genes, pathways, or compounds associated with biological traits and disease-related processes. Researchers generate or select fly populations carrying defined mutations, introduce genetic or environmental perturbations, and score observable outcomes such as development, behavior, fertility, or survival. Because Drosophila has a short generation time, well-characterized genetics, and conserved cellular pathways, screens can efficiently connect genotype with phenotype. These studies support research in development, neurobiology, aging, and disease mechanisms, while compound screens can reveal candidate therapeutics and genetic interactions for further investigation in more complex models.

RNAi Screening for Host Factors Involved in Vaccinia Virus Infection using Drosophila Cells

Novel host factors involved in viral infection can be identified through cell-based genome-wide loss of function RNAi screening. A Drosophila cell culture model is particularly amenable to this approach due to the ease and efficiency of RNAi. Here we demonstrate this technique using vaccinia virus as an example.

This video describes a screening method to identify transgenic Drosophila flies, whose genome was modified using CRISPR-Cas9. CRISPR-Cas9 is a powerful genome-editing tool that revolutionized the way scientists can manipulate an organism's genome. In the example protocol, we will see how to identify CRISPR-generated mutants, in which an insertion of a transactivation sequence replaces the first exon of the branchless (bnl) gene, thus creating a bnl gene-specific driver line, bnl-LexA.

We describe a streamlined protocol for generating "fillet" preparations of Drosophila embryos of specific genotypes. This protocol allows efficient execution of a variety of genetic screens. It also allows excellent visualization of structures in the late embryo.
