Drosophila Toxicity Testing

Drosophila toxicity testing is the use of fruit flies to evaluate how chemicals, drugs, environmental contaminants, or other stressors affect living organisms. In controlled assays, flies receive defined exposures, and researchers measure dose-dependent changes in survival, development, reproduction, behavior, or physiological responses across their life cycle. Because Drosophila shares conserved cellular and genetic pathways with other animals, these tests can reveal toxic mechanisms and provide an efficient model for comparing compounds. Applications include environmental toxicology, drug safety assessment, genetic studies of disease, and early screening that helps prioritize substances for more complex biological testing.

Liver macrophages, named Kupffer cells, are responsible for the capture of circulating nanoparticles. We describe here a method, of high cell purity and yield, for Kupffer cell isolation. The modified LDH assay is used here to measure the toxicity induced by carbon nanotubes in Kupffer cells.

Toxicity Testing in Animals

2026

Toxicity tests in animals are grounded on two main assumptions: first, the effects observed in laboratory animals can be extrapolated to humans, especially when adjusted for body surface area; second, high-dose exposure in animals is essential to identify potential human hazards from lower doses. This is based on the quantal dose-response concept, which faces the challenge of extrapolating results from relatively few test animals to much larger human populations. For example, a 0.01% incidence...

Drosophila melanogaster testes can be rapidly and efficiently isolated from adult males using dissecting needles. With practice, one can readily isolate in one or two days an amount of testes sufficient for the analysis of DNA or RNA by high throughput sequencing or more traditional molecular biology methods or of protein for antibody- or enzyme-based assays.
