Drought Tolerance Screening

Drought tolerance screening is the systematic evaluation of plants for their ability to maintain growth, function, or survival when water availability is limited. In biology, researchers impose controlled water deficits and compare genotypes using indicators such as germination, biomass, leaf water status, wilting, and recovery after rewatering. These measurements reveal differences in drought-responsive traits, including water conservation and continued physiological activity under stress. Screening supports the selection of resilient plants for breeding, helps clarify mechanisms of drought adaptation, and improves studies of how environmental stress affects plant development and productivity.

This protocol was developed to screen a large germplasm collection of the leafy vegetable lettuce (Lactuca sativa L.) for drought-tolerance, in order to identify a small candidate pool of lettuce for use in physiological, molecular, and genetic studies to identify underlying drought-tolerance traits along with breeding programs.

We describe a protocol to examine the development of opioid-induced hyperalgesia and tolerance in mice. Based on the measurement of thermal and mechanical nociceptive responses of naïve and morphine-treated animals, it allows to quantify the increase in pain sensitivity (hyperalgesia) and decrease in analgesia (tolerance) associated with chronic opiate administration.
