Epineurium Perineurium

The epineurium and perineurium are connective-tissue coverings that organize and protect peripheral nerves. The epineurium forms the tough outer sheath around an entire nerve, while the perineurium surrounds bundles of axons called fascicles; tightly joined perineurial cells also contribute to the blood-nerve barrier by regulating movement between the nerve and surrounding tissue. Together, these layers provide mechanical support, maintain a stable environment for nerve fibers, and help preserve organized signal transmission. Their structure is important in biology, histology, and clinical research involving peripheral nerve injury, inflammation, regeneration, and surgical repair.

Generating a Rat Model of Tibial Neuroma Transposition

2025

Source: Brakkee, E. M., et al. Surgery and Behavioral Testing in the Tibial Neuroma Transposition Model in Rats. J. Vis. Exp. (2023).This video demonstrates a procedure to generate a rat tibial neuroma transposition (TNT) model. After exposing the tibial nerve in the hind leg of an anesthetized rat, transect the nerve. Next, create a subcutaneous tunnel, thread the severed nerve through the tunnel, and suture it in place. In the absence of a target for reinnervation, disorganized axonal...

Patch Clamp Recordings on Intact Dorsal Root Ganglia from Adult Rats

This manuscript describes how to prepare intact dorsal root ganglia for patch clamp recordings. This preparation maintains the microenvironment for neurons and satellite glial cells, thus avoiding the phenotypic and functional changes seen using dissociated DRG neurons.

Inducing a Facial Nerve Injury in a Rat Model

This video demonstrates a surgical protocol on a rat model, detailing steps from anesthesia preparation to facial nerve injury induction. This protocol is useful for studying nerve regeneration and surgical techniques in medical research.

Construction of a Composite Regenerative Peripheral Nerve Interface in a Rat Model of Nerve Injury

Source: Svientek, S. R., et al. Fabrication of the Composite Regenerative Peripheral Nerve Interface (C-RPNI) in the Adult Rat. J. Vis. Exp. (2020).This video demonstrates the surgical construction of a composite regenerative peripheral nerve interface (C-RPNI) in a rat model of peripheral nerve injury. The procedure involves attaching a transected peroneal nerve to a muscle graft and overlaying it with a dermal skin graft to promote nerve regeneration. Over time, motor and sensory neurons...

Nerve Repair Using a Bionic Conductive Scaffold Combined with Electrical Stimulation

Take an anesthetized rat with an exposed intact right sciatic nerve, which controls leg movement and sensation.Excise a nerve segment, creating a gap, severing the neuronal axons, and disrupting signal transmission. The severed distal axons deteriorate, detaching specialized nerve-repair or Schwann cells.Suture a bionic conductive nerve scaffold, a soft tube with microchannels, to the epineurium, the nerve’s outer layer, at both cut ends.Implant paired electrodes near the nerve ends, passing...
