Equine Cloning

Equine cloning is the production of a genetically matched horse by transferring the nucleus of a somatic cell into an egg cell whose nucleus has been removed. In somatic cell nuclear transfer, electrical or chemical activation stimulates the reconstructed egg to divide into an embryo, which is implanted into a surrogate mare for gestation. The resulting foal shares the donor’s nuclear DNA, although mitochondrial DNA and developmental effects can differ. In biology and veterinary research, equine cloning supports the preservation of valuable genetics, replication of elite or sterile animals, and study of early embryonic development, while also raising important questions about efficiency, animal welfare, and genetic diversity.

A Simple Microaspiration Technique for Isolating Somatic Cells from Cryopreserved Equine Semen as Nuclear Donors for Cloning

2025

This protocol presents a simple microaspiration technique to isolate viable somatic cells (SCs) from cryopreserved equine semen. Unlike traditional approaches, this technique avoids long in vitro cultures and overcomes sperm contamination, enabling immediate use in somatic cell cloning.

Molecular Cloning

2023

Molecular cloning is a set of methods, which are used to insert recombinant DNA into a vector - a carrier of DNA molecules that will replicate recombinant DNA fragments in host organisms. The DNA fragment, which may be a gene, can be isolated from a prokaryotic or eukaryotic specimen. Following isolation of the fragment of interest, or insert, both the vector and insert must be cut with restriction enzymes and purified. The purified pieces are joined together though a technique called...

Reproductive Cloning

2019

Reproductive cloning is the process of producing a genetically identical copy—a clone—of an entire organism. While clones can be produced by splitting an early embryo—similar to what happens naturally with identical twins—cloning of adult animals is usually done by a process called somatic cell nuclear transfer (SCNT). Somatic Cell Nuclear Transfer In SCNT, an egg cell is taken from an animal and its nucleus is removed, creating an enucleated egg. Then a somatic cell—any cell that is not a sex...

Surgical Closure of Equine Abdomen, Prevention, and Management of Incisional Complications

This paper describes a method of abdominal wall closure using tension and apposition sutures in horses undergoing ventral midline laparotomies. It also describes methods for prevention and management of postoperative incisional complications, such as bandaging, negative pressure therapy, and in case of wound breakdown, the application of retention sutures.

Culturing and Harvesting of HSPC Clones: A Method to Obtain the Singly-Sorted HSPC Clones

2023

This video describes the protocol for isolating HSPCs from the bone marrow and sorting them using a fluorescent activated cell sorting technique, which is a method to obtain singly-sorted HSPCs. We culture these sorted cells to obtain HSPC clones.
