Barley Grain Diet

A barley grain diet is a feeding regimen in which barley provides a substantial portion of dietary energy and nutrients, making it useful for studying how grain composition influences biological function. Its effects arise from the digestion of starch and protein, while dietary fiber, including beta-glucan, can slow nutrient absorption and undergo fermentation by gut microorganisms. In biology and nutrition research, controlled barley-based diets help investigators examine digestion, metabolism, gut microbial activity, growth, and health outcomes in model organisms or livestock. Comparing barley with other grains can clarify how dietary components shape physiology and inform feed formulation, animal production, and human nutrition research.

Here we describe a common method to induce chronic liver injury in mice by feeding of a choline-deficient and ethionine-supplemented (CDE) diet. We demonstrate health monitoring, liver perfusion, isolation, and preservation. A time course of six weeks can inform about liver injury, pathohistology, fibrosis, inflammatory, and liver progenitor cell responses.
