Fluorescent Dextran Uptake

Fluorescent dextran uptake is a cell biology method that uses fluorescently labeled dextran to monitor fluid-phase endocytosis and intracellular transport. Cells internalize dextran from their surrounding medium through endocytic vesicles, allowing fluorescence microscopy or related measurements to reveal uptake efficiency, vesicle formation, and subsequent trafficking within the cell. This approach helps researchers assess endocytic activity, compare membrane transport under different experimental conditions, and examine how drugs, mutations, or environmental changes affect cellular uptake. Because dextran is generally biologically inert and available in different molecular sizes, fluorescent dextran uptake provides a versatile readout for studying membrane dynamics and intracellular compartmentalization.

Here, we present a method for visualizing the uptake of 3 kDa Texas Red-labeled dextran in auditory hair cells with functional mechanotransduction channels. In addition, dextrans of 3–10 kDa can be used to study endocytosis in hair and supporting cells of the organ of Corti.
