Fly Medium Preparation

Fly medium preparation is the process of making a nutrient-based culture substrate that supports the growth, development, and maintenance of laboratory flies, especially Drosophila. The method typically combines water with a gelling agent, carbohydrates, and yeast or another nutrient source; heating dissolves and sterilizes the mixture, while cooling allows it to solidify before distribution into culture vials or bottles. Properly prepared medium provides consistent food, moisture, and physical support for eggs, larvae, and adult flies. It is essential for maintaining healthy stocks, coordinating genetic crosses, conducting developmental and behavioral studies, and ensuring reproducible results in biological research.

Drosophila has emerged as a significant model system for dissecting the cellular and molecular underpinnings of behavioral responses to alcohol. Here we present a protocol for the collection of alcohol sensitivity data in a circadian context that can be easily applied to other experiments and is well-suited for undergraduate research.
