Fm 1-43 Dye Uptake

Fm 1-43 dye uptake is a fluorescence-based method for monitoring membrane trafficking in living cells, particularly endocytosis and synaptic vesicle recycling. The amphipathic styryl dye binds to the outer leaflet of the plasma membrane but becomes trapped in newly formed internal membranes when endocytic vesicles bud inward, producing a measurable increase in fluorescence; subsequent exocytosis releases the dye and reduces the signal. Researchers use this approach to visualize and quantify vesicle cycling, assess synaptic activity, and investigate how cellular conditions or molecular components regulate membrane retrieval and release. Its dynamic readout links membrane transport mechanisms with cell communication and function.
