Golgi Apparatus Staining

Golgi apparatus staining is a microscopy-based method for selectively labeling the Golgi apparatus, the organelle that modifies, sorts, and packages proteins and lipids inside eukaryotic cells. In a typical protocol, cells are fixed and permeabilized, then exposed to dyes or antibodies against Golgi markers such as GM130, followed by fluorescent detection and imaging. The resulting signal reveals Golgi ribbon organization, cisternal distribution, and changes in organelle structure. In biology, this approach supports studies of intracellular trafficking, secretion, cell polarity, and disease-associated defects, while enabling comparisons between normal, treated, and genetically altered cells.

The precise localization of Golgi residents is essential for understanding the cellular functions of the Golgi. However, conventional optical microscopy is unable to resolve the sub-Golgi structure. Here we describe the protocol for a conventional microscopy based super-resolution method to quantitatively determine the sub-Golgi localization of a protein.
