Headspace Sampling

Headspace sampling is a technique for analyzing volatile compounds by examining the gas phase above a solid, liquid, or biological sample, rather than injecting the sample itself. In a sealed vial, volatile molecules partition between the sample matrix and its headspace until they approach equilibrium; an aliquot of the gas is then withdrawn, often with a syringe or automated sampler, and introduced into gas chromatography or gas chromatography-mass spectrometry for separation and identification. In biology, this approach measures microbial or cellular volatile organic compounds, tracks metabolism, and characterizes chemical signaling while reducing matrix interference and protecting analytical instruments. It supports studies of infection, food biology, environmental samples, and physiological change.

Sampling Methods: Sample Types

2024

Sampling materials are classified into three main types: solid, liquid, and gas. Solid samples include a variety of substances, such as sediments from water bodies, soil, metals, and biological tissues. Two standard methods for extracting sediments from water bodies are grab sampling and piston coring. Grab sampling involves using a device to collect a discrete sediment sample from the bottom of a water body with minimal disturbance. Grab samples do not always represent the entire area due to...
