Hepatocyte Loss

Hepatocyte loss is the reduction or depletion of hepatocytes, the primary functional cells of the liver, and is a central feature of many forms of liver injury. It can occur when toxins, infection, oxygen deprivation, or immune activity trigger cell stress, apoptosis, or necrosis faster than the liver can replace damaged cells. Persistent loss disrupts metabolism, detoxification, and tissue organization, while incomplete repair may promote inflammation and fibrosis. Studying hepatocyte loss helps explain liver diseases, evaluate drug toxicity, and assess regenerative responses in cell culture, animal models, and emerging therapies for restoring liver function.

Isolation of Human Hepatocytes by a Two-step Collagenase Perfusion Procedure

A modified two-step collagenase perfusion procedure for isolation of human hepatocytes is described. This method can also be applied to other mammalian livers. The isolated hepatocytes are available in high yield and viability, making them a suitable model for scientific research in areas such as liver regeneration, pharmacokinetics and toxicology.

Hepatocyte-specific Ablation in Zebrafish to Study Biliary-driven Liver Regeneration

To help assess the molecular mechanisms underlying zebrafish biliary-driven liver regeneration, we established a liver injury model in which the nitroreductase-expressing hepatocytes are genetically ablated upon metronidazole treatment. In this protocol, we describe how to adeptly manipulate, monitor and analyze hepatocyte ablation and biliary-driven liver regeneration.
