Herpesvirus Infection

Herpesvirus infection occurs when a herpesvirus enters host cells, replicates, and establishes a persistent infection that can affect tissues ranging from skin and mucous membranes to the nervous system. After binding to cellular receptors, the virus delivers its genome into the nucleus, directs production of viral proteins and progeny virions, and may then enter latency, during which its genome remains in host cells with limited gene expression. Reactivation can resume viral replication when cellular or immune conditions change, producing recurrent disease or enabling transmission. Studying herpesvirus infection helps biologists investigate host–pathogen interactions, immune control, viral latency, diagnostic methods, and antiviral strategies.

(2014)This video demonstrates intranasal delivery of murine herpesvirus in mice to induce a lung-specific immune response. Lung tissue is subsequently processed to obtain cytokine-containing supernatants for downstream assessment of host immune responses.

Establishment and Quantification of De Novo Lytic Infection by Cell-free Kaposi's Sarcoma-Associated Herpesvirus

This protocol describes an in vitro system to model de novo lytic infection of Kaposi's sarcoma-associated herpesvirus using BAC16-derived virions. The method enables investigation of early viral replication and dissemination. Infectious virus production is quantified by GFP-based infectious units assays and real-time PCR of encapsidated viral genomes.

(2019)This video demonstrates the clonal isolation and PCR-based screening of recombinant herpesvirus following site-specific excision of a GFP marker. Fluorescence-activated cell sorting is used to isolate non-fluorescent infected cells, and junction PCR confirms retention of the target gene in GFP-negative clones.

We describe a protocol to identify key roles of host signaling molecules in lytic replication of a model herpesvirus, gamma herpesvirus 68 (γHV68). Utilizing genetically modified mouse strains and embryonic fibroblasts for γHV68 lytic replication, the protocol permits both phenotypic characterization and molecular interrogation of virus-host interactions in viral lytic replication.

Here, we present a protocol for the development and validation of a quantitative PCR method used for the detection and quantification of EHV-2 DNA in equine respiratory fluids. The EHV-2 qRT-PCR validation protocol involves a three-part procedure: development, characterization of qRT-PCR assay alone, and characterization of the whole analytical method.
